In a phase 2 trial, participants received 3 DNA/HIV-1 primes and a recombinant TV (rTV)/HIV-1 boost at 8 weeks (8-wk group) or 24 weeks (24-wk group) post-final prime. We investigated the vaccinia-specific immunological and overall transcriptomic responses to the rTV/HIV-1 boost. All participants in the supplemental study received three DNA/HIV-1 primes at Weeks 0, 4, and 8, followed by a single subcutaneous dose of rTV/HIV-1 (Beijing Biological Preparations Institute Co., Ltd.). Seven of the 16 participants received the rTV/HIV-1 boost 8 weeks after the last DNA prime (8-week group), whereas the other 9 participants received the rTV/HIV-1 boost 24 weeks after the last DNA prime (24-week group). Blood was drawn from each volunteer into EDTA-2K Vacutainer tubes (BD Biosciences) on the day of the rTV/HIV-1 boost (Day 0, pre-vaccination and 4 h post-vaccination) and on Days 1, 3, 7, 9, 14, 28, 90, 180, and 360 thereafter.
