Transcriptome analysis of Chx10-Cre Rb1/p107 and Chx10-Cre Rb1/p107/Hells P21 retinae

We used novel genetically engineered mouse models to investigate the role of HELLS during tumorigenesis. Loss of HELLS drastically decreased the incidence of retinoblastoma, delayed tumor progression, and increased overall survival. Retinae from Rb1/p107 DKO and Rb1/p107/Hells TKO mice at postnatal day 21 were analyzed for gene expression using RNA-seq. Retinal mRNA profiles of 21-day-old Chx10-Cre Rb1/p107 and Chx10-Cre Rb1/p107/Hells mice were generated by deep sequencing, in triplicate, using Illumina HiSeq 4000.
本研究采用新型基因工程小鼠模型,探究HELLS在肿瘤发生过程中的作用。敲除HELLS可显著降低视网膜母细胞瘤的发生率,延缓肿瘤进展,并延长整体生存期。我们对出生后第21天的Rb1/p107双敲除(DKO)小鼠与Rb1/p107/Hells三敲除(TKO)小鼠的视网膜样本,采用RNA测序(RNA-seq)开展基因表达分析。通过Illumina HiSeq 4000测序平台,对21日龄Chx10-Cre Rb1/p107及Chx10-Cre Rb1/p107/Hells小鼠的视网膜mRNA表达谱进行深度测序,实验设置三次生物学重复。
