To better understand how odorant receptor expression in female Aedes aegypti mosquitoes influences chemosensory receptor expression in the legs, we performed Illumina RNA-seq on the pooled forelegs and midlegs samples taken from wildtype animals (Orlando strain) and Orco5/16 loss of function mutant mosquitoes. For each genotype, we sequenced five biological replicates for a total of 10 samples. Samples from all tissues were barcoded and pooled in an equal ratio before distributing the pool across two lanes on a NovaSeq SP flow cell, sequenced in a single sequencing run.
