Bacteroidota are abundant members of the human gut microbiota that shape the enteric landscape by modulating host immunity and degrading dietary- and host-derived glycans. These processes can be mediated by the secretion of outer membrane vesicles (OMVs) by members of the Bacteroidota. There remains controversy regarding the physiological relevance of OMVs in the gut because our understanding of OMV biogenesis and regulation in Bacteroidota is incomplete. In this study, we performed a novel high-throughput screen to identify genes required for OMV biogenesis in Bacteroides thetaiotaomicron.
