ddRAD sequencing of Irish populations of Salvelinus alpinus

ddRAD sequencing of 288 Arctic char (Salvelinus alpinus) samples from 18 Irish populations and two outgroups (UK and Finland). DNA was extracted from fin clips, gillrakers and muscle tissue using the Wizard SV Genomic DNA Purification System (Promega, UK), following the manufacturers instructions. DNA concentration of samples was estimated using agarose gel electrophoresis and the AccuBlue Broad Range dsDNA Quantitation Kit (Biotum) on a CLARIOSTAR plate reader. Libraries were prepared using a modified version of the ddRAD protocol of Peterson et al (2012), as described in Maroso et al (2018). Libraries were sequenced on the Illumina NextSeq550 platform at the Genomics Core Technology Unit (Queen's University, Belfast) using paired-end 151bp sequencing.The associated study explored for the first time the taxonomy and the phylogeography of the Irish Arctic char using high-throughput data and will be soon available.
