We sequenced two strains that resulted from a horizontal chromosome transfer (HCT) experiment described in Ma et al. Nature 2010 (DOI: 10.1038/nature08850). A donor strain Fol007 carrying a phleomycin resistance gene (BLE, confers resistance against zeocin) inserted in chromosome 14, 180 bp downstream of the SIX1 ORF, and a recipient strains Fo47 carrying a hygromycin (HYG) resistance gene, inserted randomly into the genome, were coincubated and double-resistant colonies were selected. HCT was confirmed using CHEF-gels and Southern blots. One of the strains that obtained one transferred chromosome (chromosome 14) Fo1A and one of the strains that obtained chromosome 14 and an additional small chromosome, Fo2A, were sequenced in this study to determine whether the regions for which we observed evidence of horizontal chromosome transfer in natural isolates, correspond to chromosome 14 and the small additionally transferred chromosome in Fol007.
