Periodontitis induces myeloid lineage skewing of the bone marrow progenitor cells. In this study, we performed single cell RNA-sequencing in LSK (Lin—Sca-1+cKit+ ) cells to determine whether the periodontitis-induced myeloid lineage bias persists after the resolution of periodontal inflammation. C57BL/6 mice were trained by subjecting them to LIP for 21 days followed by ligature removal (to enable inflammation resolution) for 14 days (21dL/14dR, L) or were not ligated during the entire period (NL, N). Hematopoietic progenitors (LSKs; Lin-cKit+Sca1+) in mice bone marrow were sorted by FACS for single-cell transcriptomics analysis.
