Gene expression profile at single neuron level of mouse hippocampal neuron, sampled from ex vivo acute brain slice

Hippocampal neurons exhibit heterogeneous properties, such as physiological and molecular expression patterns. To identify the gene expression patterns related to the physiological properties, we used single-cell RNA sequencing for neurons recorded juxtacellularly in vivo. Hippocampal dorsal CA1 neurons were recorded juxtacellularly and then fluorescently labeled by electrical stimulation in vivo using the ICR mice. The brains were then isolated and sectioned. The somas of the recorded and labeled neurons were manually suctioned with a glass pipette and analyzed using Quartz-seq2.
海马神经元(Hippocampal neurons)具有异质性特性,涵盖生理特性与分子表达模式。为鉴定与神经元生理特性相关的基因表达模式,我们对经体内胞旁记录(juxtacellularly in vivo)的神经元实施了单细胞RNA测序(single-cell RNA sequencing)。本研究以ICR小鼠为实验模型,对海马背侧CA1神经元开展体内胞旁记录,随后通过体内电刺激对目标神经元进行荧光标记。后续我们分离脑组织并进行切片,通过玻璃微管手动抽吸收集已完成记录与荧光标记的神经元胞体,最终采用Quartz-seq2技术完成测序分析。
