mRNA profiles of embryonic day 14.5 wild-type (WT) and Pich-KO mice were generated by deep sequencing, in triplicate, using Illumina GAIIx. The sequence reads that passed quality filters were analyzed at the transcript isoform level with two methods: Burrow-Wheeler Aligner (BWA) followed by ANOVA (ANOVA) and TopHat followed by Cufflinks. 本研究通过Illumina GAIIx测序平台,对胚胎第14.5天的野生型(wild-type, WT)及Pich基因敲除(Pich-KO)小鼠开展三次生物学重复的深度测序,以获取其mRNA表达谱。对通过质量过滤的测序读段,我们在转录本异构体水平采用两种分析方案:其一为Burrow-Wheeler Aligner(BWA)结合方差分析(ANOVA),其二为TopHat结合Cufflinks。本数据集聚焦野生型与Pich-KO小鼠胎肝LSK细胞转录组的定量分析。
