CUT&RUN profiling to compare SOX9 and SOX10 binding in non-transfected A375 cells and A375 cells transfected with SOX9-HA. We sought to evaluate the impact of SOX9 overexpression on binding profiles in human melanoma cell culture. Using A375 or SKMel5 human melanoma cell lines, binding profiles for proteins of interest were compared between non-transfected cells and cells transfected with tagged SOX9. CUT&RUN binding profiles of up to 5 proteins-- H3K4me3, IgG, SOX9, SOX10, as well as either HA or Myc in transfected samples-- were evaluated in up to 3 (transfected)-4 (non-transfected) external replicates for A375 cells and 1 (transfected) -2 (non-transfected) external replicates for SKMel5 cells.
