Role of HDAC6-STAT3 in immunomodulatory pathways in Colorectal cancer cells

We investigate whether HDAC6 affects STAT3 activation in Colorectal Cancer cells (CCD-841 human colon normal cells, HT29 and HCT116 cells), through treatments with Nexturastat A (NexA), a specific HDAC6 inhibitor. A BRB-seq assay in dual conditions, showed a reduced expression of common genes involved in immunomodulatory pathways in CRC cells. These results suggest that treatments with specific HDAC6 inhibitor would reduce the functionality of STAT3 in CRC cells, impacting the expression of immunomodulator genes involved in the inflammatory and immune response. Therefore, the use of specific HDAC6 inhibitors may be a reasonable and an interesting adjuvant strategy for help in immunotherapy, since HDAC6 inhibitors would decrease the expression of different immunomodulators of immune response
