In this study, we successfully overexpressed and characterized the α-L-rhamnosidase gene BtRha from Bacteroides thetaiotaomicron VPI-5482 in E. coli. As compared on the enzyme properties, BtRha was very rare to have the highly selective to hydrolyze the α-1,2 and α-1,6 glycoside linkages between rhamnoside and rhamnoside, rhamnoside and glycoside, respectively. And the protein was more efficient and selective in biotransformation epimedin C to icariin. Thus, this study provides a useful α-L-rhamnosidase, which may be used to improve the enzymatic conversion of epimedin C to icariin.
