The effect of amplicon length on Taxonomic Analysis of Oxford Nanopore Sequencing. Optimization of ONT Sequencing on meta-genomics samples

Only fifteen percent of the bacteria have a single 16s rRNA region. Not only this phenomenon but also the Outer Membrane vesicles (OMVs) are affecting the meta-genomic diversity analysis. Outer membranes possess proteins, RNAs and DNA fragments. The existence of Ribosomal subunits either Protein or RNA counterparts is shown by various researchers. Genomes size restriction of bacteria has brought up various adaptions tackling with environmental changes. Population genomics is bigger than the total bacterial genomic size by the help of various extrachromosomal nucleic acids. The extrachromosomal DNAs are also detected in OMVs and even whole translation machinery can be also detected in OMVs. The DNA fragments inside OMVs might also make an analysis bias with rRNA subunits. Our goal is to optimize the taxonomic amplicon size to capture good resolution of bacterial diversity where the enlisted problems won't make any problem.
