Despite the function of Myc oncogene has been extensively studied, little is known concerning its role in Natural killer (NK) cells. By performing RNA-sequencing analyses comparing Myc-deficient to Myc-sufficient NK cells, we aim at determining the biological function of Myc in these innate lymphocytes. Overall design: To this aim we generated Ncr1cre Mycfl/fl mice, in which Myc expression is deleted in NK cells at the immature stage. We sorted the immature CD27SP NK (sorted as NK1.1+CD27+CD11b-CD3/19-) cells from the bone marrow of the Ncr1cre Mycwt/wt (WT) or Ncr1cre Mycfl/fl (KO) mice and subjected them to RNA-sequencing analysis. Ncr1cre Mycfl/fl and Ncr1cre Mycwt/wt mice were generated by crossing Ncr1cre and Mycfl/fl mice which were previously described (Narni-Mancinelli et al., 2011, 10.1073/pnas.1112064108; Trumpp et al., 2001, 10.1038/414768a). All mice were backcrossed multiple times on a C57BL/6 background.
