Transcriptomics profiling of singleton and twin placentas in the absence or presence of preeclampsia.

Twin pregnancies incur a higher risk for preeclampsia (PE) than singletons, but the pathogenesis might differ. To better understand this, we performed RNA sequencing on 32 individual dichorionic (D) and monochorionic (M) twin placental samples grouped by clinical PE diagnosis and 10 newly sequenced singleton (S) PE placentas. We performed RNA sequencing (RNA-seq) using 42 human placental villous tissue samples grouped as follows: (1) singleton (S) PE placental samples with a range of clinical PE disease severities (n=10); and (2) 32 individual villous placenta samples from 16 diamniotic twin pregnancies classified by chorionicity (dichorionic, D; and monochorionic, M) and PE diagnosis: No PE D (n=7 pregnancies), No PE M (n=2 pregnancies), PE D (n=5 pregnancies), and PE M (n=2 pregnancies). In addition, 3 new samples from placentas used in our prior study (corresponding to S06 [GSM6175121], S18 [GSM6175133], and S20 [GSM6175135] from GSE203507) were extracted and prepared using the paired-end library and sequencing protocol to assess how this compared to the original sequencing protocol (single-end, 50 bp reads).
