Expression of immune response and tissue repair genes in human colonic CD4s upon cytokine or TCR stimulation.

Human Colonic CD4 T cells consist of several subsets differing in their expression of CD161. We exposed these cells to either cytokine, TCR or combined stimulation and assessed the subsequent changes in gene and protein expression with an emphasis on cytokine receptors and gene associated with tissue repair. Overall design: Lamina propria mononuclear cells (LPMCs) from three donors (141, 194 and 267) were isolated and stimulated tih IL12/18 (C), plate-bound aCD3 (T) or a combination of both (TC) overnight. Cells were stained with BD Abseq antibodies and BD Sample Tags, CD4+ T cells were sorted, pooled and loaded onto a BD Rhapsody cartidge for single cell capture followed by cDNA cythesis and library preparation.
