We studied the effects of loss of hepatic O-GlcNAc Transferase (OGT), the enzyme that attaches the O-GlcNAc to protein, in a choline-deficient and ethionine (CDE) supplemented model. We placed hepatocyte-specific OGT knockout mice (OGT-KO) and controls on the CDE diet for 2 weeks. We then performed RNA sequencing. We then repeated the 2-week CDE diet with the addition of a 7-day normal diet recovery period. We then performed RNA-Sequencing on isolated RNA from whole liver lysate to determine differentially expressed genes. Overall design: Comparative gene expression profiling analysis of RNA-seq data for control and OGT-KO mice (equally pooled RNA from n of 3) with no recovery and a 7 day recovery after CDE diet. cDNA was sequenced using 2 lanes targeting 25 million reads/sample.
