Pseudomonas aeruginosa PAO1 was used as a model in this study. It is well know that infection caused by this opportunistic pathogen is very hard to be treated due to its intrinsic and acquired resistance mechanisms. We aim to apply phage therapy in combination with antibiotics to treat P. aeruginosa PAO1. Therefore, lytic phage were isolated from influent collected from wastewater treatment plant using Pseudomonas aeruginosa PAO1 as host. Influent collected from wastewater treatment plant was condensed by Polyethylene Glycol (6000) after the removal of solid particle by centrifugation. Phages were isolated by double layer agar method, whereby single plaque was picked up and purified and propagated. Propagation of phages was conducted by using P.aeruginosa PAO1.
