Streptomyces malaysiensis DSM4137 total RNAseq

Streptomyces malaysiensis DSM4137 has been cultured in the 10ml of TSBY medium for up to 60 hours, samples for RNA extraction were taken at 12h, 18h, 30h, 48h, 60h.For RNA extraction, 1 mL of sample was mixed with twice the amount of RNAprotect Bacteria Reagent (QIAGEN, 76506), vortexed for 5 sec, incubated for 5 min at room temperature and then centrifuged at 10 min at 5000 x g. The supernatant was carefully decanted and dried by dabbing on a paper towel. The resultant pellet was flash-frozen in liquid nitrogen and then stored at -80 until needed. Preferably, RNA was extracted from samples immediately.Illumina RNAseq library was prepared using TruSeq stranded LT RNA prep kit, and sequenced on the MiSeq instrument (2x76bp).
