Independent first-principles rediscovery of BRCA1-PARP1 and BRCA1-POLQ synthetic lethality on a 12-node Boolean network in 728 ms

A 12-node hand-coded Boolean network of the DNA double-strand-break repair pathway, scanned with a generic pair-knockout brute-force attractor enumerator, rediscovers PARP and POLQ as the canonical synthetic-lethal partners of BRCA1 loss in 728 ms of total wall time, with no machine learning, no training data, and no prior knowledge of which partners to test. PARP is the FDA-approved BRCA1-SL drug-class target (olaparib, niraparib, rucaparib, talazoparib); POLQ is the next-generation BRCA-SL approach in active phase 1/2 trials.
