Microarray expression profile and bioinformatics analysis of circular RNAs in the phenotypic switching of rat vascular smooth muscle cells: a preliminary study

In order to further study the role of circular RNA in the phenotypic transformation of vascular smooth muscle cells (VSMCs), the differential expression profile of circRNA in the phenotypic transition of VSMCs induced by platelet-derived growth factor-BB (PDGF-BB) was screened using chip technology. Vascular smooth muscle cells from rat thoracic aorta were induced with 20ng/ml PDGF-BB as the experimental group and compared with the control group. After induction for 24 hours, the differentially expressed circRNA was screened by circular RNA chip.

为深入探究环状RNA(circular RNA, circRNA)在血管平滑肌细胞(vascular smooth muscle cells, VSMCs)表型转化中的作用,本研究采用芯片技术筛选了血小板衍生生长因子-BB(platelet-derived growth factor-BB, PDGF-BB)诱导的VSMCs表型转化过程中差异表达的circRNA表达谱。 实验以大鼠胸主动脉来源的血管平滑肌细胞为研究对象,将经20ng/ml浓度的PDGF-BB诱导处理的细胞设为实验组,并与未诱导的对照组进行对比。 诱导处理24小时后,通过环状RNA芯片筛选得到差异表达的circRNA。
