Single cell RNA sequencing of human primary glioblastoma

Glioblastoma (GBM) is the most lethal form of central nervous system cancer with a median survival of less than 2 years. Here, we examined the single-cell transcriptomes of primary GBM excised tumors from five different patients to explore the underlying mechanisms of GBM development. Overall design: We performed the single-cell RNA sequencing of primary GBM excised tumors from five different patients. Tumour specimens were washed by PBS and mechanically cut into small pieces using sterile surgical scissors, and then digested into single cell suspensions. Single-cell RNA libraries were constructed and suquenced using Chromium Next GEM Single Cell 5' V2 GEM, Library & Gel Bead Kit (10×Genomics) and Illumina NovaSeq 6000.
