The multifaceted roles of the DNA polymerase subunit Pol32 in maintaining genome integrity

A total of 40 diploid WYpol32 strains that spontaneously produced red and white colonies were analyzed using a chromosome IV microarray. The diploid strain WYpol32 carries a homozygous ade2-1 mutation, which is partially suppressed by the SUP4-o gene on chromosome IV. A break between the centromere and the SUP4-o insertion site can induce recombination, leading to crossover events and the formation of red-white sectored monoclonal colonies. Analyzing 40 pairs of red-white sectors can reveal the critical role of the Pol32 in recombination.
