In this study, the post-transcriptional role of chloroplast RNA-binding protein CP29A was investigated in Arabidopsis thaliana and Nicotiana tabacaum. NGS-based techniques were used to determine the in vivo binding sites and the in vitro binding preference of CP29A, using an eCLIP-based and RNA-Bind-N-Seq protocol, respectively. Based on these binding data the role of CP29A for the expression rbcL, one of the main targets, was in analyzed in greater detail using a series of reverse genetic and molecular biolgy tools. The data sets for the eCLIP-based and RNA-Bind-N-Seq experiments are part of this submission, and also include binding data for the chloroplast RNA-binding protein CP33B, which served as an additional control for the specficity of the binding assays.
