In-vivo transcriptome analysis was performed to understand the Mtb adaptation strategies to survive in the host and establish infection in BTB. Gene expression profiling of Mtb within abscesses or necrotic specimens obtained from patients with BTB TB compared to exponentially growing in-vitro Mtb culture using whole genome microarray was studied. Transcriptional profile of mycobacteria within abscess or necrotic tissue from BTB TB patients in comparison to exponentially growing Mtb was assessed. Two controls (Mtb H37Rv) and six clinical specimens from microbiologically confirmed bone TB patients were analyzed. One sample out of six, was excluded from the data analysis because of involvement of joints as all other five clinical specimen were from Spinal TB patients. Agilent M.tb expression array, 8*15K (Design ID: 026323) designed by genotypic technologies Pvt. Ltd. Was utilised for the study.
