Characterizing the effect of long-term cigarette smoke exposure on bronchospheres gene expression Overall design: Bronchospheres were differentiated in 384-well plates on 50% Matrigel for two weeks, after which they were exposed to cigarette smoke extract (CSE) at the indicated percentages (diluted in HBEC differentiation media) for 7, 14, and 21 Days. Then they were lysed directly in the plate with Qiagen buffer RLT and then RNA was extracted from the cells using a Qiagen RNeasy kit with the elution step done in water.
