DNA methylation in mouse cancer model with ketogenic diet and mifepristone treatment

DNA methyaltion analysis using Illumina Infinium Mouse Methylation Beadchips in different groups of mice with and without exposure to medroxyprogesterone acetate and 7,12-dimethylbenzanthracene MPA/DMBA (which induces mammary gland tumours), with and without mifepristone (an antiprogestin) treatment, and with and without a ketogenic diet. A total of 86 mice are organised into three experimental sets. The discovery set consists of 9 mice without exposure to MPA/DMBA and 9 mice with MPA/DMBA exposure (of which 8 developed a mammary gland tumour). These mice were on a normal diet and were not treated with mifepristone. Tissue samples from normal mammary gland, mammary tumor, cervix, oviduct, blood, adipose tissue, kidney, liver, lung, and spleen were collected from both groups. The intervention set conists of of 35 mice fed with normal diet which were randomized into four groups: without MPA/DMBA exposure and without mifepristone, without MPA/DMBA exposure and with mifepristone, with MPA/DMBA exposure and without mifepristone, and with MPA/DMBA exposure and with mifepristone. Tissue samples from normal mammary gland, mammary tumours, cervix, oviduct and blood were taken from all subgroups. The validation set conists of of 33 mice fed with ketogenic diet which were randomized into four groups: without MPA/DMBA exposure and without mifepristone, without MPA/DMBA exposure and with mifepristone, with MPA/DMBA exposure and without mifepristone, and with MPA/DMBA exposure and with mifepristone. Tissue samples from normal mammary gland, mammary tumours, cervix, oviduct and blood were taken from all subgroups.
