Characterization of the meningeal leukocytes in 3xTg-AD mice

We characterize the meningeal leukocyte (CD45+ cells) in wild-type and 3xTg-AD mice, a transgenic model of Alzheimer's disease (AD). Overall design: Pools of 8 female mice (3xTg-AD or wild-type) were used for single-cell RNA-Seq (scRNAseq) analysis. Leukocytes isolated from the meninges were washed with PBS, and labeled with an anti CD45–BV480 antibody (Becton Dickinson). The cells were re-suspended in PBS supplemented with 10% FBS), and viable CD45HIGH cells were sorted on a FACSAria Fusion (Becton Dickinson). Both the 3xTg-AD and wild-type samples consisted of > 98% viable cells.
