Prokaryotic microbial communities in various biofilter carriers from Japanese drinking water treatment plants (DWTPs) were analyzed using full-length 16S rRNA gene (16S rDNA) amplicon sequencing. Samples included sand from a slow sand filter (SA) and biological activated carbon (BAC) from ozone-BAC filters (BACFs) and up-flow biological contact filters (U-BCFs). Both DNA extracts and cDNA PCR products (reverse-transcribed from RNA) were sequenced to capture total and potentially active microbial communities, respectively. The study compared microbial communities across different carriers and examined the effects of location, filter type, and nucleic acid type (DNA vs. RNA). Sample names include abbreviations for filter type (SA, BACF, UBCF), followed by a number indicating different biological filters when available, and DWTP location codes (TK, CB, HG, OS, FO).
