Determination of mechanical strength of stemDetermination of stem flexural strength: In the mature stage, the stems of the wild type and mutant bc21 were cut from the second internode, cut into 10 cm, and placed on a support 7.5 cm apart. During the measurement, the stems were parallel to the ground, a digital display push-tension meter was placed in the middle of the stem, and moved vertically downward until the stem broke off. The peak pressure was read and measured for 3 repetitions.Measurement of stem tensile force: Take 10 stems from the second internode of the mature mutant bc21 and the wild type, and cut them into 10 cm in length. The stems are perpendicular to the ground. Use a universal mechanical testing machine to clamp the two ends of the stems.2. Determination of cell wall componentsThe same internodes were cut off from the stems of wild type and mutant bc21 at the mature stage of cultivation in the field, and the leaf sheaths were peeled off and put into clean sealing bags. Each material was measured for 3 times and repeated. The samples were sent to Suzhou Grisi Biotechnology Co., Ltd. to determine the content of cellulose, hemicellulose and lignin in the stem cell wall, and the results were analyzed.
