Begomovirus are major problem destructive to cucurbit crop cause economic losses both quality losses and substantial yield. This virus is widely spread and can readily transmit by whitefly. Genetic resistance of plant are good and most effective method to reduce loss occurred. The aim of this study is evaluate cucurbit resistance to Squash leaf curl Yunnan virus isolate PK-B12 (SLCuYV-PK-B12) under greenhouse condition. To study the method for screening of cucurbits to begomovirus by multiplied of virus in susceptible plant. To determine the appropriate time for detected of virus. The result shown that 4 week after transmission is the best time for test. After that evaluation of 152 accessions of cucurbits include wax gourd 73 accessions, 72 accessions of pumpkin, and 7 accessions of ridge gourd were screened for resistance to this virus. Virus transmission by whitefly vector. Detected of viruses from samples with enzyme-linked immunosorbent assay (ELISA). Disease rating based on percent of infected plants classified accessions Resistant (R), moderate resistant (MR), susceptible (S) and Highly Susceptible (HR). The results of this evaluation the plants showed disease reaction resistant of 28 accessions in wax gourd (Benincasa hipsida) 16 accessions, 5 accessions of pumpkin (Cucurbita moschata) and 7 accessions of ridge gourd (Luffa acutangula). These species will be used to improve the resistance to SLCuYV-PK-B12.
