The G Protein-Coupled Receptor ADGRG6 Maintains Mouse Growth Plate Homeostasis by Negatively Regulating IHH Signaling

ADGRG6 is a cartilage-enriched G protein-coupled receptor (GPCR). Using molecular mouse genetics and spatial transcriptomics approaches, we demonstrated that Adgrg6 has a vital role in regulating chondrocyte differentiation and growth plate homeostasis by positively regulating the formation and/or maintenance of the PTHrP (+) cell population and negatively regulating the IHH signaling in postnatal growth plates. Spatial Transcriptomics (Visium, 10x Genomics) was performed on Hindlimbs of P20 Col2Cre; Adgrg6f/f mutants and Cre (-) littermate (wild-type) controls

ADGRG6是一种富集于软骨组织的G蛋白偶联受体(G protein-coupled receptor,GPCR)。本研究借助小鼠分子遗传学与空间转录组学(Spatial Transcriptomics)技术手段,证实Adgrg6可通过正向调控出生后生长板中甲状旁腺激素相关蛋白(Parathyroid Hormone-related Protein,PTHrP)阳性细胞群的形成与/或维持,同时负向调控印度豪猪信号通路(Indian Hedgehog,IHH),在软骨细胞分化及生长板稳态维持过程中发挥关键作用。本研究采用10x Genomics公司的Visium空间转录组平台,对出生后20天(P20)的Col2Cre;Adgrg6f/f突变小鼠及其Cre阴性同窝野生型对照小鼠的后肢组织开展了空间转录组测序。
