Here we addressed the mRNA transcriptome of Long-Term Hematopoietic Stem Cells (LT-HSC) and Multipotent Progenitor (MPP) -3 and -4 purified from WT and Helios KO mice. By analyzing the differentially expressed genes among WT and KO condition we found that Helios deletion affects transcription mostly in the LT-HSC compartment and favor a megakaryocyte mRNA lineage priming at the expense of the lymphoid one. Overall design: Long-term hematopoietic stem cells (LT-HSC) and multipotent progenitors myeloid and lymphoid biased (MPP-3 and -4 respectively) isolated from the bone marrow from 3 WT and 3 Helios KO mice by flow cytometry. Total RNA was extracted from LT-HSC, MPP3 and MPP4 cells and used to prepared libraries that were sequenced. The analysis of the differentially expressed genes in WT and KO cell populations revealed that Helios deletion favor a megakaryocyte lineage priming mRNA profiling at the expense of the lymphoid one in these cells.
