Aging is a major risk factor for chronic kidney disease. Aged mice (21 months old) exhibited enriched transcriptomic signatures of fibrosis and cellular senescence in their kidneys compared to young mice (4 months old), and dietary supplementation with nicotinamide (NAM) was insufficient to reverse these disease signatures. Overall design: 3 groups of 6 (young) or 8 (old, old+NAM) animals. Kidney tissue from 4 months old (young) and 21 months old (old) wild-type (WT) C57BL/6JRj male mice. Some old animals supplemented with nicotinamide (NAM) mixed with the diet at a dose of 200 mg/kg bw/day, 1.6g/kg food for 14 weeks.
