The transcriptional coactivator ANGUSTIFOLIA 3 (AN3) stimulates cell proliferation during Arabidopsis leaf development, but the molecular mechanism is largely unknown. We aimed to identify direct targets of AN3 by analyzing the presence of AN3 at their genomic regions. Thereto, Tandem Chromatin Affinity Purification (TChAP), an alternative technique to isolate chromatin complexes (A. Verkest, T. Abeel, C. Lanz, E. Van De Slijke, J. Van Leene, N. De Winne, Y. Van de Peer, F. Van Breusegem, D. Inzé, K. Vandepoele, and G. De Jaeger; in preparation), was performed on Arabidopsis cell culture overexpressing HBH-tagged AN3 and followed by sequencing. Furthermore, AN3 binding to the genomic regions of important transcriptional regulators, shown to be rapidly differentially expressed after induction of AN3 activity, will be analysed.
