Spindlin1 is a histone reader with three Tudor-like domains and its reader activity of histone methylation could be attenuated by SPINDOC. To validate such a role in cellular context, we performed ChIP-Seq in HEK293 cell with SPINDOC overexpressed cell line and control cell line. Overall design: Flag-Spindlin1 stably expressed HEK293 cells were transiently transfected with HA-SPINDOC. Essentially, cells were crosslinked with formaldehyde. After chromatin sonification and centrifugation, the supernatant was aliquoted and subjected to immunoprecipitation using ChIP-grade Flag beads, 4 µg of H3K4me3 and H3K9me3 antibody respectively. Immune complexes were purified by Protein A/G agarose. After reverse crosslinking, the resultant DNA was eluted and purified for sequencing.
