Nascent strand sequencing (NS-seq) in U2OS cells without or with NFIB depletion to identify de facto activated replication origins. Overall design: U2OS cells were infected with lentiviruses carrying control shRNA or NFIB shRNA. RNA-primed nascent DNA at replication origins (0.5-2.0 kb in length) were purified and subsequently sequenced using the Illumina platform.
