Lorlatinib is an oral medication used to treat a specific type of lung cancer called ALK-positive non-small cell lung cancer (NSCLC). Lung cancer is generally divided into two main types: non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC). NSCLC is the most common, accounting for about 85% of all lung cancer cases. Anaplastic lymphoma kinase (ALK)-positive lung cancer is a rare form of NSCLC caused by an abnormal fusion between the ALK gene and another gene. This fusion leads to continuous signals that cause uncontrolled cell growth, which can result in cancer. Lorlatinib has been shown to be very effective compared to previous treatments, with many patients living longer without their disease worsening. However, a significant number of patients taking lorlatinib experience weight gain, which can impact their overall health and quality of life. The weight gain is often linked to changes in the fat distribution; where fat is stored in the body. This can increase the risk of developing other health problems, such as high cholesterol, diabetes and high blood pressure. Currently, the exact cause of this weight gain is not fully understood, nor is there a clear way to manage it effectively. This study aims to better understand how lorlatinib causes weight gain in lung cancer patients. We will monitor changes in body weight and use CT scans (a common imaging test) to measure changes in fat distribution. Specifically, we will focus on fat around the organs in the abdomen (visceral fat) and fat just under the skin (subcutaneous fat). These specific changes are important as they are linked to higher health risks. Second, we aim to better understand the effects of weight gain in lung cancer patients by assessing their quality of life and mood. By analyzing data from patients treated with lorlatinib, we hope to identify patterns of weight gain and changes in fat distribution. This research will help us better understand this side effect and lead to better recommendations for preventing or managing weight gain. These recommendations may include changes in lifestyle, diet, or even additional medical treatments. Ultimately, this study could help improve the long-term health and quality of life for patients receiving lorlatinib.
