This study applied Oxford Nanopore Technology (ONT) long-read sequencing of full-length 16S rRNA amplicons from natural soil amended with lignocellulosic biomass and from microbial consortia enriched on carboxymethyl cellulose (CMC)-based substrates. To reduce barcoding costs, a hybrid workflow combining PacBio ligation-based barcodes for sample indexing with ONT Ligation Sequencing Kit library preparation was used, providing a scalable and cost-effective approach for high-resolution microbiome profiling.
