Real-time pH curves were monitored during silage production from grass, alfalfa, and maize. Samples were taken before ensiling, during the phase of the fastest pH decline, after the pH curve had flattened, and after two months, which is a representative period for complete ensiling. These were then analyzed using the long-read sequencing platform offered by Pacific Biosciences. This study was the first to combine microbiome analyses with real-time pH monitoring during ensiling. This enables direct correlation of the microbiome with the actual ensiling phase, which provides more accurate information about the microorganisms responsible for the main pH drecrease. This information is relevant for the production of new and effective inoculants.
