Mycobacteria-induced secretion of cytokines and chemokines in healthcare workers with apparent resistance to infection with <i>Mycobacterium tuberculosis</i>.

Cytokines and chemokines in PBMC supernatants (stimulated with Mtb and BCG), plasma and supernatants from Quantiferon-TB PLUS assay (Nil and TB1 stimulated) after incubation for 16-20 hours. Concentrations were measured by Luminex using the ProcartaPlex Human Monitoring Panel 65-plex panel (Invitrogen™) according to manufacturers' instructions. Biological samples were collected from individuals with latent TB infection (LTBI, IGRA+ &amp; TST+), people with resistance to Mtb infection (Resisters, IGRA- &amp; TST-) and people with previous but successfully treated TB. IGRA+ for IFN-gamma level &gt;0.35IU/mL; IGRA- for IFN-gamma &lt;0.35IU/mL; TST+ for induration &gt;10mm and TST- for induration &lt;10mm

本数据集包含经结核分枝杆菌（Mycobacterium tuberculosis, Mtb）与卡介苗（Bacillus Calmette-Guérin, BCG）刺激后的外周血单个核细胞（Peripheral Blood Mononuclear Cell, PBMC）上清液、血浆，以及经16-20小时孵育后的Quantiferon-TB PLUS检测试剂盒（分为空白对照Nil组与结核抗原TB1刺激组）所得上清液中的细胞因子与趋化因子；采用Luminex平台结合ProcartaPlex Human Monitoring Panel 65-plex检测面板（Invitrogen™品牌），依照制造商说明书完成浓度测定。生物样本采集自三类人群：潜伏性结核感染（Latent Tuberculosis Infection, LTBI）个体，即干扰素γ释放试验（Interferon-Gamma Release Assay, IGRA）阳性且结核菌素皮肤试验（Tuberculin Skin Test, TST）阳性者；结核分枝杆菌感染抵抗者（Resisters），即IGRA阴性且TST阴性者；以及既往感染结核分枝杆菌且已成功治愈的肺结核患者。本研究的判定标准为：IGRA阳性定义为干扰素γ（Interferon-gamma, IFN-γ）水平＞0.35IU/mL，IGRA阴性定义为IFN-γ水平＜0.35IU/mL；TST阳性定义为皮肤硬结直径＞10mm，TST阴性定义为皮肤硬结直径＜10mm
