Epigenetic effects of vitamin C on bone and osteogenic differentiation by analyzed RNA-seq, hMeDIP-seq and ChIP-Seq analysis Femurs from 20 week old Gulo-/- mice or 11 week old Prrx1-Cre driven Tet1fl/fl, Tet2fl/fl or Tet1fl/fl and Tet2 fl/fl mice were dissected, bone marrow was flushed, periost was removed and remaining bone tissue was thorogly washed in ice cold PBS. MC3T3-E1 cells were collected at day 7 or day 28 of osteogenic differentiation. For all samples RNA, DNA or chromatin was extracted using commercially available extraction kits and/or standard protocols.
