This project explores the optimization of an amplicon-based whole genome sequencing approach using Unique Molecular Identifiers (UMIs) to improve the accuracy of Oxford Nanopore Technologies (ONT) long-read sequencing. The method was applied to mock microbial community samples and human blood cultures spiked with common bloodstream infection pathogens. We observed total error rates below 0.1% with the V9 sequencing chemistry, and further improvement to below 0.05% with the V14 chemistry.
