Single-Cell Barcoded Lineage Tracing using KP4 PDA Cell Line - Pre and Post Drug Treatment with Trametinib

The KP4 PDA cell line (Untreated, DMSO-Treated and Trametinib-Treated) was used to experimentally assess plasticity between MAPK-Positive and MAPK-Negative states of Pancreatic Malignant Cells Overall design: Cells were transduced with an average of ~3 million random 27-nucleotide barcodes and were subjected to single-cell RNA-Seq at two different time points, named as T1 and T2 (10 population doublings). Cells were either Untreated, treated with 0.15% DMSO for 24h or treated with Trametinib in 0.15% DMSO for 24h.
