Gene expression of immune cells in a lung cancer mouse model

We used in situ vaccination with autologous dendritic cell secreting CCL21, in combination with immune checkpoint inhibitor, anti-PD-1, to treat oncogene-driven non-small cell lung cancer in a murine model. The single cell transcriptome of the treated cancer tissues were measured. Cells were stained with Zombie-NIR LIVE/DEAD stain and CD45 antibody for 20 min a 4oC. Viable leukocytes were sorted using BD Biosceinces Aria II cell sorter with 100-mm nozzle. The 10X Genomics platform (10X Genomics, Pleasanton, CA) was utilized for assessing single-cell transcriptomic. In brief, sorted immune cells were loaded into 10X Genomics Chromium microfluidic chips with 3’ v3 chemistry to generate single-cell gelbead emulsions (GEMs).
