Type 2 inflammation contributes to the pathology of airway diseases such as asthma and eosinophilic pneumonia. Yet how sensory neurons innervating the airways respond to type 2 airway inflammation is poorly understood. The goal of this study was to characterize transcriptional changes in sensory ganglia innervating the lungs in the setting of type 2 airway inflammation induced by Alternaria allergen treatment. 8–10-week-old C57BL6 mice with the Jak1 gene conditionally deleted in Nav1.8-expressing neurons (homozygous for a floxed mouse Jak1 allele and crossed to Nav1.8-Cre = del-Jak1) or Cre-negative littermates (LM) were treated with 2 ug of Alternaria extract daily for four days to induce asthma-like airway inflammation. At day 5, nodose ganglia and thoracic dorsal root ganglia were harvested and the tissue was dissociated to yield single nuclei suspensions for downstream single-nuclei sequencing.
