Evaluating Pulmonary Toxicity of PFOS and its Alternative OBS Using Spheroids of A549 Cells

Transcriptome analysis of OBS on spheroids of A549 cells. including 4 replicates of SC group and 4 replicates of high concentration group.Briefly, after 7 days of exposure, A549 cell spheroids from per replicate in both the SC and OBS-H groups were respectively collected for RNA sequencing. Total RNA was extracted using Trizol Reagent (Invitrogen, USA), and its concentration and purity were assessed using a Nanodrop2000 spectrophotometer (Thermo Fisher Scientific, USA). RNA integrity was verified through agarose gel electrophoresis. RNA-sequencing was performed on an Illumina HiSeq 4000 SBS Kit (300 cycles) platform at Shanghai Majorbio Bio-Pharm Technology Co., Ltd. (Shanghai, China).
