Soil Biome Variation of Lupinus nipomensis in Wet-Cool vs. Dry-Warm Microhabitats and Greenhouse Conditions

We used the CALeDNA methods to generate multilocus metabarcoding libraries targeting the ITS1 (fungal DNA), CO1 (invertebrates and protists), 18S (eukaryotes), and 16S (bacterial and archaeal) regions. Soil samples were collected from greenhouse-grown Lupinus nipomensis individuals and from experimental field plots representing wet-cool and dry-warm microhabitats. DNA was extracted using the Qiagen DNeasy PowerSoil kit, amplified with Nextera-adapted primers, and sequenced on an Illumina MiSeq with a target depth of 50,000 reads per sample per metabarcode. Lupinus nipomensis is an endangered species in California and is federally listed.
