To understand the role of 3D chromatin architecture in establishing tissue-specific expression of the CFTR gene, we mapped chromatin interactions using Circular Chromosome Conformation Capture (4C) and epigenomic data such as H3K27ac and Assay for Transposase-Accessible Chromatin (ATAC-seq) on Capan-1 pancreatic cells. Overall design: We used 4C to detect chromatin interactions with the promoter of the CFTR gene in Capan-1 cells. Caco-2 and HepG2 cells were used as controls for 4C interaction mapping.
