Natural rubber is cis-polyisoprene, a polymer formed by sequential condensation of isopentenyl disphosphate (IDP) moieties by rubber transferase (Cornish, 2001). There are two pathways for IDP anabolism - one in the cytosol, using mevalonate, and one in the plastid, using 1-deoxy-D-xylulose 5-phosphate/2-C-methyl-D-erythritol 4-phosphate. One approach to increase rubber production is to engineer the metabolic flux through the plastid. This requires knowledge of the plastid genome sequence.
